You could, but you won't get boot screen f.ex.
Mac Pro owners have earlier only had the option to either use a hacked card, or officially either a GTX285 or a Quadro card (which costs like $1500 and upwards).
Now they can get a new compatible card for like a third of the cost of a Quadro.
Here is a "worst case scenario" for AMD/ATi users for pro use:

You will actually use a full work day (436 minutes= 7 hours and 16 minutes) to render something on an AMD card that you could do in minutes on a NVidia card
I guess if you live on using After Effects, you can afford the $100 premium for a mac branded version

Happened to me, twice.
First megaupload shut down, and I had all my ports there.
Then mediafire did delete my files, for some unknown reason.
I lost some ports each time, but the popular ones is never hard to get a hold of again.
But, I don't give up that easy.

Pros with normal wine is that it will take less space.
Cons is that you won't get a self-contained app, you won't have a GUI.
You will also need to make different prefixes for each app that need a different configuration.
Also you are using wine, and you are a part of the thriving wine community.
Wineskin is just a tool that puts wine into a GUI with it's own x server, you are using the same wine.
